<p>近年电动汽车市场增长迅速，采用电动汽车已被视为减少交通工具温室气体排放的关键因素。电动汽车的关键部件是锂离子电池(LIB)。然而，锂离子电池排放的温室气体最多可占电动汽车排放总量的三分之一。因此，目前正在进行大量研究，以提高锂离子电池的比能以及从废旧锂离子电池中回收金属，从而改善电动汽车的总体成本和对环境的影响。</p>



<p>在电池研发当中，溶解度是设计和优化回收工艺的一个重要特性。而结晶是一种有效的分离和提纯方法，可用于回收纯净的无机材料，从而实现重复使用并减少对原材料的需求。</p>

<p><strong><meta charset="utf-8"><strong><span class="has-inline-color has-vivid-cyan-blue-color">应用案例：有色电池盐</span></strong></strong></p>



<p><strong>方法：</strong>Technobis 专业的结晶研究仪器 &#8211; Crystalline 是使用热变量法有效收集和分析溶解度数据的理想工具。将粉末状水合盐称量到 8 ml 结晶瓶中，然后加入 3 ml 去离子水。样品配有顶置钩形搅拌器。样品以 0.1 ºC/min 的速度加热至 70 ºC，然后保持 10 分钟以达到平衡，再以 0.1 ºC/min 的速度冷却至 5 ºC。利用 Crystalline 高分辨率相机和新型粒子检测技术，可以在有色和较暗的样品中采集清点和浊点，这在以前是一项挑战。但利用这项技术，我们能够收集硫酸铜和硫酸镍的溶解度和析出曲线。</p>

<p><strong>结果：</strong>收集了五水硫酸铜的多个清点和浊点，从而绘制出溶解度和 MSZW 曲线。在很宽的温度范围内，溶解度逐渐增加。研究发现，MSZW 的宽度非常窄，约为 8ºC。这对扩大规模生产特定晶型提出了挑战。利用高分辨率照相机，我们能够捕捉到自发成核和随后的晶体生长过程，从而获得金字塔形晶体。</p>

<p>收集了六水硫酸镍的几个浓度清点，并绘制了溶解度曲线。还检测到了浊点，但数据点并不匹配。通过图像分析，我们可以观察到两种不同的形态：针状和金字塔状。据观察，不同的硫酸镍水合物具有不同的晶体形态。研究发现，温度低于 31.5ºC时，硫酸镍会以正交七水合物形式结晶，高于 31.5ºC时，则会以四水合物/六水合物形式结晶。因此，不同的结晶行为可归因于硫酸镍形成了不同的水合形式。</p>

<p>2022伊始，国家药监局药审中心发布《化学仿制药晶型研究技术指导原则（试行）》，对仿制药晶型选择、晶型研究思路、研发中晶型问题的关注点、药物晶型的表征和控制等作出明确的技术规范指导，并于发布当日实施执行。</p>

<p>该指导原则指出研究原料药的多晶型现象以及晶型可能对药物全生命周期各环节产生的影响，是保证药物质量的核心内容和关键环节，对化学仿制药的研发具有重大意义，明确了仿制药晶型研究过程中的关注点：</p>

<h2><strong><span class="has-inline-color has-vivid-cyan-blue-color">自动化方案，结晶研究速度质量双提升</span></strong></h2>



<p>《化学仿制药晶型研究技术指导原则（试行）》结合我国仿制药晶型研究的现状并参考国外监管机构相关指导原则而起草，它对国内晶型研究进一步规范，它的发布对国内晶型研究无疑具有重大意义。作为从业者，如何在其指导下，高效开展晶型研究工作，以实现药品的安全性、有效性和质量可控性？</p>

<p>强生实验室使用 Technobis Crystal16 测定了苄星邻氯青霉素在各种溶剂中的溶解度。实验采用了温度变化法，按程序进行升降温，并实时检测、记录体系的溶解性数据，<strong>14 小时内即可完成 4 种不同溶剂下的溶解度测定，而测试所需的API少于500毫克</strong>，真正实现降本增效。</p>

<p>荷兰代尔夫特理工大学 (Delft University of Technology)使用了 Technobis Crystalline 对甘露醇进行结晶工艺优化，考察升降温速率、添加剂、加晶种、反溶剂对生成晶型的影响。通过 Crystalline 在线晶粒观测模块以及其他多种功能，实验人员可以监控结晶过程，掌握结晶机理，快速找到适合于优势晶型的工艺条件。</p>

<p>上海药物研究所应用 Chemspeed SWING Crystal 高通量结晶工作站自动完成液体及固体加样，同时进行<strong> 192 个平行结晶实验</strong>，完成多种工作流程：多晶型、盐、共晶、溶解度筛选等，系统地考察各种结晶条件：如溶剂、浓度、蒸发速率、温度、降温速率和搅拌速度等。</p>

<h2><span class="has-inline-color has-vivid-cyan-blue-color">01 溶剂对 API 产品和工艺的影响</span></h2>



<p>在药物开发过程中，结晶工艺贯穿于整个 API 的分离与合成。其中，结晶工艺开发中溶剂的选择至关重要，它影响产品的化学纯度、固态形式(晶型)、固态属性 (颗粒属性)。</p>

<p><strong><span class="has-inline-color has-vivid-cyan-blue-color">对化学纯度的影响</span></strong></p>



<p>根据 ICH 指导原则，溶剂有其 “天然属性”，分为三类。当我们选择某种溶剂时，已经确定了它在产品中的最高残留限。</p>



<p>通过溶液结晶可以除杂，但如果溶剂使用不当，也会产生不需要的杂质。例如，在常见的磺酸体系中，如果用醇做溶剂，可能会产生磺酸酯杂质。因此，一开始选择溶剂的时候，需要充分考虑它对产品化学纯度的影响。</p>

<p>一些 API 在特定的溶剂中可以形成溶剂合物，可能对开发无水晶型和水合物造成干扰。</p>



<p>在开发某一个 API 的结晶工艺时，首先需要确定目标晶型。有些 API 存在不止一种晶型，通过梳理晶型转化关系图，可以避开干扰溶剂 (可以形成溶剂化合物的溶剂)，缩小溶剂筛选范围。</p>



<p>针对水合物的开发，需要研究它与无水晶型或水合物的关键水活度，从而确定溶剂中合适的水含量范围。对于无水晶型的开发，除了考虑水合物的影响外，还需关注目标晶型与其他无水晶型的关系：单变还是互变，从而确定合适的结晶温度。</p>



<p><strong><span class="has-inline-color has-vivid-cyan-blue-color">对物料固体属性的影响</span></strong></p>



<p>溶剂是影响晶体形貌的重要因素之一。</p>



<p>通过晶体生长实验，可以观察到不同种类的溶剂会对晶体的大小、形貌产生影响。有时候溶剂比例的变化，也会对晶体形貌产生影响。</p>

<p><strong><span class="has-inline-color has-vivid-cyan-blue-color">案例一：颗粒控制</span></strong></p>



<p><strong>背景</strong></p>



<p>已有实验筛选数据，晶型转化关系研究充分，目的改善颗粒大小，提高产品的堆密度。</p>



<p><strong>筛选溶剂的流程</strong></p>



<ul><li><sub>针对目标晶型，前期根据晶型关系转化图，避开不需要的溶剂。</sub></li><li><sub>根据溶解度结果，筛选出 8 种溶剂，进行晶体生长评估，从中确定 4 种候选溶剂。</sub></li><li><sub>针对 4 种候选溶剂，进行溶液稳定性评估，结合溶剂种类 (2 种 2 类和 3 类溶剂)，最终优先选择了 2 种 3 类溶剂。</sub></li><li><sub>针对候选的 2 种 3 类溶剂，进一步做了溶解度和结晶实验，最终选择了 1 种更经济、环保的溶剂。</sub></li><li><sub>确定完溶剂体系后，进行结晶工艺开发和优化。</sub><br></li></ul>

<p><strong><span class="has-inline-color has-vivid-cyan-blue-color">案例二：晶型控制</span></strong></p>



<p><strong>背景</strong></p>



<p>某化合物，前期已经发现了 11 种晶型，目标是开发其中一种无水晶型的结晶工艺。前期研究数据有限，没有晶型关系转化图，为溶剂筛选增加了难度。</p>



<p><strong>筛选溶剂的流程</strong></p>



<ul><li><sub>对常用的 2 类和 3 类溶剂，进行溶解度测定，得到正溶剂、反溶剂信息，再进行晶型稳定性实验。<br></sub></li><li><sub>设计了反溶剂添加实验，但没有发现目标晶型，实验陷入困境。</sub></li><li><sub>通过前期研究资料，了解到目标晶型是在乙腈体系制备得到。同时发现乙腈体系湿品为一种新晶型，晾干后转为目标晶型，推测目标晶型为 desolvate。该晶型与乙腈有溶剂依赖性，选择乙腈为溶剂。</sub></li><li><sub>若项目开发时已知目标晶型的晶型属性，在后续溶剂选择中可以直接确定乙腈，不需要进行溶剂筛选工作，可以大大缩短项目周期，节省成本。</sub></li><li><sub>应用 <strong><a href="https://www.nikyang.com/benchtop-solutions/technobis/crystal16/">Technobis Crystal16 </a></strong>, 很快地测定了在乙腈体系下的溶解度曲线和亚稳态区 (mszw)。</sub></li><li><sub>根据获得的实验结果，设计了降温结晶实验，理想地获得了目标晶型。</sub></li></ul>

<p>溶剂的选择对 API 结晶工艺开发至关重要，对产品的纯度、晶型、形貌、工艺的收率、体积效率皆有重大影响。在选择溶剂时，<strong>如何降低残留？如何保证目标晶型及溶剂的稳定性？如何自动测定溶解度？</strong></p>



<p>在力扬企业与苏州晶云药物科技共同举办的<strong>《结晶工艺开发中的溶剂选择》</strong>网络会议上，小伙伴们留下了他们在结晶工艺开发中遇到的问题，晶云药物高级研究员余姝老师进行了详细解答，以下内容供大家学习参考。</p>



<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>如何降低产品中的溶剂残留？</strong></p>



<p>选择溶剂时，考虑 ICH 对溶剂的分类，优先选择溶残限值比较高的溶剂，有利于降低控制溶残的难度；避免形成溶剂合物导致溶残超标；控制晶体生长和析晶速度，使晶体有充分的时间生长，避免溶剂包裹。</p>



<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>湿品和干品是否每种化合物都需要做稳定性 PXRD (API 在烘干前后)？</strong></p>



<p>目标晶型在所选溶剂下的稳定性以及溶剂的稳定性是一个需要优先考虑的问题。因此，API 的多晶型转化关系尽可能在药物开发早期研究，可以帮助确定优势晶型及工艺开发中的溶剂体系，判断某些体系中是否干品和湿品存在不同晶型。若前期研究不充分，需要在结晶工艺开发中，在确定溶剂体系及干燥条件后考虑多晶型及转变的风险。</p>



<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>已知产物可以以结晶态存在，纯度还不错，如果产物结晶容易成油，可以从哪几方面考虑?</strong></p>



<p>这种情况需要在工艺设计中加晶种控制，加入目标晶型的晶种是一个非常有效的手段，但前提必须明确所选晶型在当前体系中能够稳定存在。此外，考虑原料纯度的影响。</p>



<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>如果遇到几乎什么溶剂都不溶的化合物，升温至80℃ 的溶解度也是微溶，需要如何处理？</strong></p>



<p>考虑提高温度和二元溶剂。使用高温时需要确定化合物的在高温溶剂中的稳定性，如果热稳定性好，可以考察更高温度，极端案例的工艺温度可以提升到了100℃ 以上；单一溶剂无法溶解的，可考虑使用二元溶剂。</p>



<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>水活度是什么？如何测定水活度？如何根据水活度指导结晶工艺？</strong></p>



<p>水活度是指水在这个溶剂里面的分压，如某溶剂的水活度是 0.2，可以理解为这个溶剂对应的湿度是 20%，像水活度为 1 (100%) 即表示在纯水条件下。</p>



<p>有专门的软件计算不同溶剂不同含水量对应的水活度的关系。这个值可用于无水晶型和水合物的转换关系研究。如果化合物没有水合物的干扰，那么水活度就是非必要考虑因素。</p>



<p>可以理解为水活度只是一种潜在的可能干扰因素，看其是否是影响无水晶型和水合物的关键转化点。</p>



<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>目标晶型的确定，需要参考哪些因素或者参数？</strong></p>



<p>晶型确定主要是基于前期的多晶型筛选。根据溶解度或生物利用度的需求，先确定选盐或是游离态；再根据晶型筛选和相互转化关系确定，通常会选择热力学稳定的晶型。少数情况，可能由于溶解度等因素会选择亚稳晶型。</p>



<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>测定溶解度是不是采用 HPLC 类似含量的快速检测，看峰面积来折算溶解度？</strong></p>



<p>溶解度的检测方式有很多种，HPLC 检测是一个比较常用的方法，其优点是检测灵敏度相对较高，检测结果相对准确；但它的缺点是耗时，每个点一般需平衡 24 小时，再分离固体和液体，再稀释制备样品，然后才能上机检测，整个流程历时比较长。</p>



<p>另一种方式，主要针对无机物。比如说氯化钠，配制成饱和溶液，然后挥干称重。但目前主要做的都是有机化合物，有紫外吸收的居多，所以很少采用称重法。</p>



<p>此外，就是采用<strong><a href="https://www.nikyang.com/benchtop-solutions/technobis/crystal16/">Technobis Crystal16</a></strong>的温度变化的动态法进行溶解度曲线的自动测定，可以<strong>快速得到化合物的溶解度曲线</strong>。</p>

<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>溶解度在多少可以算反溶剂了？</strong></p>



<p>取决于和正溶剂的差异，差异越大，反溶剂的效果可能会更好，一般选用<strong>&lt; 5 mg/mL</strong>。</p>



<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>如果PXRD的图形类似，用马尔文测粒径也类似，但是API复溶时间差别很大，有什么好的方法来调节复溶时间?</strong></p>



<p>复溶时间与晶型、粒径、形貌都有关系。通过XRPD判断是不是同一种晶型，还是某个晶型与无定形的混合物或是无定形。如果是混合物，需要确定两者比例在不同批次是否是一致。</p>



<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>Crystal16是否可以自动进行不同溶剂的溶解度测定？溶解度曲线和亚稳区能自动绘制吗？</strong></p>

<p>这两个问题的答案都是肯定的。</p>



<p>针对不同的溶剂，<strong>Crystal16 单次试验可以快速平行测定最多 4 种不同溶剂的溶解度曲线</strong>。其测定原理是温度变化法，第一步，每种溶剂下配置 4 个不同浓度的过饱和样品；第二步，设置好升/降温程序，点击运行，系统便会自动监测和记录数据；第三步，系统软件中查看和处理数据，即可得到该物质在不同溶剂中的溶解度曲线或超溶解度曲线。在系统自带的数据处理软件中，会根据实验过程中检测到数据进行溶解度曲线和亚稳区的自动绘制。</p>



<p>当然，如果对于作图还有其他形式的要求，也可以将原数据导出，在 excel 或 origin 等其他制图软件中进行绘制。</p>

<p class="has-pale-cyan-blue-background-color has-background"><strong>Q</strong>. <strong>如何缩短过滤时间？</strong></p>



<p><strong>通过改善颗粒的大小和形状，保证颗粒的均一性可以缩短过滤时间</strong>。一般来说，三维生长的颗粒（如块状）优于二维生长的颗粒(片状) 优于一维生长的颗粒(针状)。颗粒在 10 微米以上，一般情况过滤还可以。</p>
