<p>流通池法是进行缓控释制剂、难溶性药物和诸多特殊剂型（如混悬剂、软胶囊、植入物、微球和脂质体）溶出度测试的优选装置。尽管多年以前药典收载了流池法设备，但没有官方的性能验证测试（PVT）方法。在这项研究中，水杨酸片被用于开发流池法的 PVT 方法。与转篮法和桨法溶蚀和零级释放机制相似，水杨酸片被证明是流池法溶出仪装置 PVT 的一个良好的潜在参考标准。</p>

<p>在实验阶段 I 中，采用实验设计法（DoE）系统研究了四个参数对水杨酸溶出度的影响。其中，片剂加载方式是影响溶出度的最重要参数；流速和流通池内径（ID）也有显著影响；而温度对溶出的影响可以忽略不计。在阶段 II 中，两个测试实验室的四个不同分析员在不同的流通池设备（即四名合作者）进行了重复性和再现性评估，并确定 PVT 的初步验收标准。第&nbsp;II&nbsp;阶段的实验条件是将片剂放置在片剂支架上，流通池内径为 12 mm 装载了玻璃珠，流速为 16 mL/min，温度为 37°C，并在 90 分钟时收集样品。PVT 的再现性通过来自第五个合作者的数据得到证实。</p>

<p>纳米混悬液给药系统是一种新型的给药系统。将药物做成纳米混悬剂有利于增加药物溶出度和提高生物利用度。对于难溶性药物，做成纳米混悬剂，可改善其体内药物动力学行为，提高药效。</p>



<p>不同工艺下的纳米混悬剂，受其处方成分差异、API粒径分布、晶型等影响，溶出行为可能有差异。</p>

<p>因此，溶出实验也是纳米混悬剂研究的重要手段。那么，不同制剂工艺下得到的不同批次纳米混悬剂，我们该如何测试它们的溶出行为是否有明显差异？溶出度测试应该如何进行？</p>



<p>从混悬剂溶出实验的角度看，使用桨法，直接将混悬剂投到溶出杯进行溶出实验，似乎是可行的方案。的确，桨法也是混悬制剂中比较常用的方法，但当中可能存在一些问题，影响实验结果，下面列举一个例子：</p>



<p></p>



<p><strong><span class="has-inline-color has-vivid-cyan-blue-color">桨</span></strong><span class="has-inline-color has-vivid-cyan-blue-color"><strong>法 </strong>&#8211; 用移液枪移取一定体积的混悬剂，加入溶出杯中</span></p>

<p>在桨法的溶出度实验下，纳米混悬剂的<strong>样品释放速度过快，无法比较不同批次样品的溶出差异</strong>。仔细思考桨法的溶出实验过程，我们可以找出当中的问题：</p>



<ul><li>移液枪移取纳米混悬剂，6个平行样品，加样的时候6杯的加样有时间差，样品的溶出在混悬剂与溶出介质接触时已经开始；</li></ul>



<ul><li>纳米混悬剂加入到溶出杯时的高度、位置可能出现差异，影响溶出结果；</li></ul>



<ul><li>小体积的纳米混悬剂一下子加入到比较大体积的溶出介质中，可能瞬间已完全溶出；</li></ul>



<ul><li>纳米混悬剂分散在溶出介质中，可能在取样时，把没溶出的样品取走，影响溶出结果。</li></ul>



<p></p>



<p><span class="has-inline-color has-vivid-cyan-blue-color"><strong>流池法 </strong>&#8211; 混悬剂被载入流通池，以闭环系统完成实验</span></p>



<p>除了桨法，当然我们可以考虑使用流池法进行实验。流池法是《中华人民共和国药典2020版》新增的方法，因其灵活性和适用范围广泛，在混悬剂的溶出研究中也适用。而<strong>流池法的闭环系统，尤其适用于缓慢释放的样品</strong>，让样品慢慢溶出。</p>

<p>从流池法的结果中看出，纳米混悬剂缓慢溶出，不同的纳米混悬剂溶出有差异。相比于桨法，流池法有以下优势：</p>



<ul><li>纳米混悬剂被载入流通池中，加样位置相对固定；</li></ul>



<ul><li>纳米混悬剂在实验开始后才与溶出介质接触，才开始溶出的过程；</li></ul>



<ul><li>纳米混悬剂可以被滤膜阻挡在流通池中，溶出过程在流通池中发生；</li></ul>



<ul><li>流通池池子体积较小，并且在溶出原理上其动力更温和，能得到较缓慢的溶出曲线。</li></ul>

<p>一篇发布在Molecular Pharmaceutics的文献[1]，也利用流池法进行了纳米混悬剂的体外释放度研究。在溶出过程中，<strong>纳米混悬剂被装载在流通池中的，已溶出的API可以透过滤膜流出流通池，而没有溶出的API则被阻挡在流通池里继续溶出</strong>。</p>

<p>对流池法方法进行优化后，可以进行不同样品的溶出实验，有效对比溶出行为的差异。</p>

<p>从以上可以看到，流池法在混悬剂的溶出度研究中有一定的优势。以流池法进行纳米混悬剂的溶出度实验，可以更真实地反映混悬剂的释放行为，提高实验结果的可靠性。如果在做混悬剂的溶出实验上碰到问题了，可以尝试一下流池法，或许有意外的收获。</p>
